ABSTRACT

Abstract BACKGROUND: Because normal male sexual differentiation is more complex than normal female sexual differentiation, there are more cases of disorders of sex development (DSDs) with 46,XY karyotype that have unclear etiology. However, Leydig and Sertoli cell markers are rarely used in distinguishing such individuals. OBJECTIVES: To evaluate the function of Leydig and Sertoli cells in individuals with genital ambiguity, 46,XY karyotype, palpable gonads and normal testosterone secretion. STUDY DESIGN AND SETTING: Case-control study with 77 patients, including eight with partial androgen insensitivity syndrome, eight with 5α-reductase deficiency type 2 (5ARD2) and 19 with idiopathic 46,XY DSD, and 42 healthy controls, from the Interdisciplinary Study Group for Sex Determination and Differentiation (GIEDDS), at the State University of Campinas (UNICAMP), Campinas, Brazil. METHODS: Baseline levels of gonadotropins, anti-Müllerian hormone (AMH), inhibin B, insulin-like 3 (INSL3), testosterone and dihydrotestosterone in cases, and AMH, inhibin B, and INSL3 levels in controls, were assessed. RESULTS: There was no significant difference in age between cases and controls (P = 0.595). AMH and inhibin B levels were significantly lower in cases than in controls (P = 0.031 and P < 0.001, respectively). INSL3 levels were significantly higher in cases than in controls (P = 0.003). Inhibin B levels were lower in 5ARD2 patients (P = 0.045) and idiopathic patients (P = 0.001), in separate comparisons with the controls. CONCLUSION: According to our findings, we can speculate that inhibin B levels may be used to differentiate among DSD cases.

ABSTRACT

In birds, male embryo the gonads develop bilateral testes, in which both left and right sides produce functional spermatozoa, whereas female embryo, only the left gonad develops into a functional ovary. Estrogen plays a key role in avian sex determination in both sexes by binding to the estrogen receptor (ER). Surprisingly, chicken estrogen receptor (cER) mRNA is expressed in both sexes; moreover; its expression is only expressed in the left male gonad. The present study aimed to localize ER protein in the left gonad of male quail embryo using immunohistochemistry. The 8-day-old male quail embryos whose embryonic sex distinguished by gonadal morphology were studied. Histology of the left male gonad displayed thin cortex containing 1 to 2 layers of the germinal epithelium, while testicular cords were observed in the medulla. ER-immunoreactive cells were only found in the germinal epithelium but not in the medulla. Localization of ER was detected in the nucleus and cytoplasm of the germinal epithelial cells. The number of ER-immunoreactive cells counted in upper, lateral, and lower regions of the germinal epithelium was 18.20±1.892, 17.60±1.887, and 16.20±1.290, respectively. This study shows the first evidence for expression of ER protein in the left male gonad of the avian embryo, indicating that ER plays a role in avian gonadal sex differentiation.


En las aves, la gónada embrionaria en los machos se desarrolla bilateralmente, ambos testículos producen espermatozoides funcionales, mientras que en el embrión hembra, sólo la gónada izquierda se convierte en un ovario funcional. El estrógeno juega un papel clave en la determinación del sexo aviar, en ambos sexos, mediante la unión al receptor de estrógeno (RE). Fuertemente los receptores de estrógenos de pollo (cRE) el ARNm se expresan en ambos sexos; además, su expresión sólo se produce en la gónada izquierda del macho. El objetivo fue localizar proteínas del RE en la gónada izquierda de embriones de codorniz macho mediante inmunohistoquímica. Se estudiaron embriones de codorniz machos a los 8 días de edad, cuyo sexo embrionario se distinguió por la morfología de las gónadas. La histología de la gónada izquierda estuvo representada por la corteza delgada que contiene de 1 a 2 capas del epitelio germinal, mientras que se observaron cordones testiculares en la médula. El RE se encontró en células inmunorreactivas del epitelio germinal, pero no en la médula. Se detectó la localización de RE en el núcleo y el citoplasma de las células epiteliales germinales. El número de células RE-inmunorreactivas en las regiones superior, lateral e inferior del epitelio germinal fue de 18,20±1,892, 17,60±1,887 y 16,20±1,290, respectivamente. Este estudio muestra la primera evidencia de expresión de la proteína de RE en la gónada izquierda del embrión aviar macho, lo que indica que el RE desempeña un papel en la diferenciación sexual de la gónada aviar.

ABSTRACT

Schistosomes are trematode parasites and of worldwide medical importance for humans and animals. Growth and development of these parasites require a specific host environment, but also permanent communication processes between the two genders. Accumulating molecular evidence indicates that the responsible interactions are mediated by signal transduction processes. Conserved signaling molecules were identified, and first approaches made for their characterization. However, no representative of the conserved family of cGMP-dependent protein kinases (cGKs) has been described in this parasite yet. Within the Schistosoma mansoni genome data-set we identified cGK homologs, of which one was investigated in more detail in this study. We present the cloning of SmcGK1, whose sequence shows homology to cGKs of higher eukaryotes. SmcGK1 was found to be gender-independently transcribed in adult schistosomes. The occurrence of SmcGK1 sense and antisense transcripts suggests that the expression of this gene is controlled at the post-transcriptional level. In situ hybridization experiments demonstrated a gonad-preferential expression profile in both genders indicating a role of SmcGK1, at least during sexual development of schistosomes. Using a cGK-specific inhibitor to treat adult schistosomes in vitro finally resulted in a multifaceted phenotype including slow motion, oocyte congestion, and reduced egg production.


Esquistossomos são parasitas trematodos de importância médica em todo o mundo para o homem e os animais. O crescimento e o desenvolvimento destes parasitas requerem um ambiente específico do hospedeiro, mas também um processo de comunicação permanente entre parasitas dos dois sexos. Evidência molecular tem se acumulado e indica que as interações são mediadas por processos de transdução de sinal. Moléculas sinalizadoras conservadas foram identificadas, e as primeiras abordagens têm sido feitas para sua caracterização. Contudo, não foi ainda descrito nenhum representante da família conservada das proteína-quinases dependentes de cGMP (cGKs) neste parasita. Analisando o genoma do Schistosoma mansoni nós identificamos homólogos de cGK, dos quais um foi investigado em mais detalhe no presente estudo. Aqui apresentamos a clonagem do gene SmcGK1, cuja sequência mostra homologia com cGKs de eucariotos superiores. Smc- GK1 foi detectada como sendo transcrita de forma gêneroindependente em esquistossomos adultos. A ocorrência de transcritos de SmcGK1 senso e antisenso sugere que a expressão deste gene é controlada em nível pos-transcricional. Experimentos de hibridização in situ demonstraram uma expressão preferencial nas gônadas em ambos os gêneros, indicando um papel para SmcGK1, pelo menos durante o desenvolvimento de esquistossomos. Usando um inibidor específico de cGK para tratamento de esquistossomos adultos in vitro finalmente resultou em um fenótipo multifacetado, incluindo movimentos lentos, congestão dos oócitos, e redução da produção de ovos.

Foram avaliadas as alterações no metabolismo materno durante a prenhez em ratas Wistar, prenhes e não-prenhes, submetidas à restrição protéica, que receberam dietas isocalóricas (15,74 kJ/g), controle ou hipoprotéica (17 por cento versus 6 por cento), distribuídas em quatro grupos (n = 7), quais sejam: controle não-prenhe (CNP) e prenhe (CP) e hipoprotéico não-prenhe (HNP) e prenhe (HP), do 1º ao 18º dia de prenhez. Parâmetros bioquímicos, hormonais e relacionados à síntese de lipídios foram considerados. Utilizou-se ANOVA a duas vias seguido de teste Tukey-HSD e teste t de Student, significância de p < 0,05. A restrição protéica elevou a síntese de lipídios e a atividade da enzima málica (EM) no fígado (FIG) e reduziu a massa ( por cento) e a razão lipí+dio/glicogênio nesse tecido, bem como reduziu a ingestão protéica (total e por cento), o conteúdo ( por cento) de lipídios na glândula mamária (GMA), as proteínas e a albumina séricas, com consequente redução nas massas da placenta e fetos. A prenhez reduziu a proteinemia, a albuminemia, a síntese de lipídios, a atividade da EM, os lipídios e o glicogênio no FIG. Mas elevou a massa corporal final, a massa ( por cento) do tecido adiposo gonadal (GON), do FIG e da GMA, e reduziu a massa ( por cento) da carcaça (CARC), a síntese e o conteúdo de lipídios no GON e, na GMA, o conteúdo de lipídios. A insulinemia elevou-se na prenhez, com glicemia reduzida, caracterizando resistência hormonal. A leptina e a prolactina também se elevaram na prenhez, sendo o aumento maior no HP. A restrição protéica na prenhez modificou o metabolismo materno, alterando a síntese de lipídios no FIG e o perfil hormonal, além de reduzir a massa da placenta e dos fetos.


Metabolism alterations were evaluated in female Wistar rats (dams) during pregnancy. Pregnant and non-pregnant dams submitted to protein restriction, were fed isocaloric (15.74 kJ/g), control or hypoproteic (17 percent vs. 6 percent) diets, and distributed in four Groups (n=7) as follows: non-pregnant control (NPC), pregnant control (PC), non-pregnant hypoproteic (NPH), and pregnant hypoproteic (PH); from Day 1 to Day 18 of pregnancy. Biochemical, hormonal and metabolic parameters related to lipid synthesis were assessed. The two-way ANOVA, followed by Tukey-HSD and Student-t tests were used, with a significance of p< 0.05. Protein restriction elevated lipid synthesis and malic enzyme (ME) activity in the liver, and reduced mass and the lipid/glycogen ratio in this tissue; it also lowered protein ingestion (total and percent), lipid content ( percent) in the mammary gland (MAG), serum proteins and albumin, with consequent reduction of placenta and fetal masses. Pregnancy reduced serum protein and albumin concentrations, lipid synthesis, ME activity, hepatic lipid and glycogen content. However, it increased final body mass; increased relative masses of gonad (GON), liver and MAG; but reduced lipid synthesis and content of GON, lipid content of MAG and the relative mass of carcass. Pregnancy Insulinemia increased during pregnancy with reduced glycemia, characterizing hormonal resistance. Leptin and prolactin were also increased during pregnancy, being the highest increase in observed in HP rats. Protein restriction in pregnancy modified maternal metabolism, altering lipid synthesis in the liver and hormonal profile and decreasing the placenta and fetus masses.

Ovarian development in Labeo dyocheilus was assessed during active reproductive phase under ambient environmental conditions in captivity and wild. Increasing day length and water temperature seemed favourable for ovarian development in female L. dyocheilus under both conditions. Gonadosomatic index (GSI) was lower in May and higher in July in captivity (6.168 and 13.366) and wild (5.798 and 16.166) respectively Ovarian development started little bit in advance in captivity with late yolk vesicle stage oocyte in May when yolk globule stage oocytes were prominent in wild. Fully grown oocytes with germinal vesicle migration (GVM) and germinal vesicle breakdown (GVBD) stages were observed in July in both conditions. Degree of transformation of developing oogonials into advance stage oocytes was observed to be better in wild fish compared to captivity reared ones. The histophysiological changes in liver corresponded well to the progression of ovarian development. Successive granulation and vacuolization of hepatocyte cytoplasm were indicative of augmented synthetic activity and probably mobilization of energy content for oocyte growth. These observations indicated that normal ovarian development of L. dyocheilus under captivity in Tarai region of Uttarakhand would be useful for success of its seed production in captivity for stock augmentation in wild or species diversification in aquaculture.

ABSTRACT

Mitotic chromosomes of the freshwater snail Pomacea patula catemacensis (Baker 1922) were analyzed on gill tissue of specimens from the type locality (Lake Catemaco, Mexico). The diploid number of chromosomes is 2n = 26, including nine metacentric and four submetacentric pairs, therefore, the fundamental number is FN = 52. No sex chromosomes could be identified. The same chromosome number and morphology were already reported for P. flagellata, i.e., the other species of the genus living in Mexico. The basic haploid number for family Ampullariidae was reported to be n = 14 in the literaure; so, its reduction to n = 13 is probably an apomorphy of the Mexican Pomacea snails. Lanistes bolteni, from Egypt, also shows n = 13, but its karyotype is much more asymmetrical, and seems to have evolved independently from P. flagellata and P. patula catemacencis. The nominotypical subspecies, P. patula patula (Reeve 1856), is a poorly known taxon, whose original locality is unknown. A taxonomical account is presented here, and a Mexican origin postulated as the most parsimonious hypothesis.

ABSTRACT

As dosagens de testosterona, sulfato de deidroepiandrosterona (SDHEA) e cortisol no líquido amniótico (LA) permitem avaliar as funçöes das adrenais e das gônadas fetais. Estudamos LAs de 41 gestaçöes, obtidos por amniocentese no segundo trimestre da gestaçäo. Trinta e sete destas gestaçöes (das quais quatro de risco para hiperplasia adrenal congênita por deficiência da 21-hidroxilase) resultaram em fetos normais, três foram anencéfalos e um, portador de deficiência da 21-hidroxilase. Os esteróides foram dosados utilizando-se radioimunoensaio previamente descritos para dosagens séricas. Valores médios (ñ DP) destes esteróides no LA de gestaçöes com fetos normais foram testosterona: 76 ñ 34 ng/del; SDHEA: 22 ñ 9 ng/ml; cortisol 19 ñ 8 ng/ml. LAs de gestaçöes que resultaram em anencéfalos apresentaram concentraçöes de SDGEA e cortisol significativamente mais baixas em relaçäo aos normais. Testosterona foi mais baixa em fetos femininos quando comparados aos masculinos normais e discretamente mais elevada que o normal no feto feminino afetado.Näo houve diferença no valor destes esteróides ao compararmos LAs de fetos de risco para a deficiência da 21-hidroxilase com os normais. Assim, a dosagem destes esteróides näo contribui de modo definitivo para o diagnóstcio de defeitos de síntese adrenal ou de anencefalia. Também näo permitem definir com segurança o sexo fetal
